Et cum spiritu tuo: cum takes the ablative
The response everyone knows is a preposition lesson: cum takes the ablative, so spiritus becomes spiritu and tuus follows it to tuo. Every time you answer the celebrant you are declining a second-declension noun correctly, which is more than most first-year students manage under exam conditions.
