is with the negative built in, "if ... not, unless", and the difference from is worth having: negates the WHOLE condition, while negates one particular word inside it. The verb behaves as it would in any conditional, indicative for an open case and subjunctive for an imagined or contrary-to-fact one.
After a negative main clause it also serves as a plain "except, but": is "nothing but", is "nobody except". English uses two different words there; Latin gets away with one because the negative in front does half the work.
